In order to explore the features which control the [4Fe-4S] cluster attachment site, the cysteine residues which are involved in ligation to the cluster have been altered to either alanine or serine. The gene for ISP-2 was cloned by PCR, and the cDNA was sequenced. The ISP-2 cDNA was subcloned into a baculovirus expression vector, pAcSG2, and insect cells were infected with the recombinant virus. Overexpressed ISP-2 was purified to homogeneity by affinity chromatography. The site-specific mutations in ISP-2 were introduced by PCR using oligonucleotide primers containing the codon for the desired amino acid change and the enzyme BsmBI. The alterations were confirmed by DNA sequencing and the proteins were expressed, purified and characterized. These proteins were fully active in the electron transfer reactions tested. The [4Fe-4S] cluster was identified as Fe-S clusters by absorption spectroscopy and electron paramagnetic resonance.
